Reward and Drugs of Abuse

چکیده
Although a number of perspectives on drug addiction exist, this chapter focuses on drug addiction as a type of pathology of learning and memory systems, a viewpoint that has become increasingly common since the mid1990s. Both natural rewards and drugs of abuse appear to use the same systems within the brain to influence and reinforce behavior, and it is well known that these systems are involved in learning and memory, particularly in connecting motivations and memories with behaviors. The next section of the chapter provides an introduction to the major issues of reward and addiction. The following three sections examine drug addiction in a manner based loosely on the stages of memory — namely, acquisition, consolidation, and storage/ retrieval. Although we shall see that drug addiction does not mirror these stages perfectly, this model provides a starting point for understanding how learning and memory, reward, and addiction interact at a neurobiological level.

Commentary: New View on Treatment of Drug Dependence
In the 1960s, discovery of pleasure system (defined as reward system) in the brain that may underlie drug reward and addiction encouraged many scientists to investigate the mechanisms by which drug abuse affects central nervous system function. In this regard, investigators developed several drugs targeting the brain reward system for drug dependence therapy. However, no positive results obtain...
متن کاملMaternal Separation and the Risk of Drug Abuse in Later Life
Maternal separation (MS) is defined as the termination of the continuity of mother-child relationship after the relationship is established. Although MS and maternal deprivation are different in terms of their definitions, these two terms are usually used interchangeably. This review aims to investigate the effect of MS on drug intake in adulthood. It has been proved that animal models are help...
متن کاملThe role of the aversive effects of drugs in self-administration: assessing the balance of reward and aversion in drug-taking behavior.
Since the first experimental demonstration that a drug of abuse supports instrumental behavior, drugs have been discussed in the context of their rewarding effects, which are assumed to drive and maintain drug-taking behavior. Indeed, drug reward has been fundamental in the formulation of most models of drug use, abuse, and addiction. Over the last several decades, however, drugs of abuse have ...
متن کاملThe Sensitivity of the Crayfish Reward System to Mammalian Drugs of Abuse
The idea that addiction occurs when the brain is not able to differentiate whether specific reward circuits were triggered by adaptive natural rewards or falsely activated by addictive drugs exist in several models of drug addiction. The suitability of crayfish (Orconectes rusticus) for drug addiction research arises from developmental variation of growth, life span, reproduction, behavior and ...
متن کاملStriatal Signal Transduction and Drug Addiction
Drug addiction is a severe neuropsychiatric disorder characterized by loss of control over motivated behavior. The need for effective treatments mandates a greater understanding of the causes and identification of new therapeutic targets for drug development. Drugs of abuse subjugate normal reward-related behavior to uncontrollable drug-seeking and -taking. Contributions of brain reward circuit...
متن کاملAddiction is a Reward Deficit and Stress Surfeit Disorder
Drug addiction can be defined by a three-stage cycle - binge/intoxication, withdrawal/negative affect, and preoccupation/anticipation - that involves allostatic changes in the brain reward and stress systems. Two primary sources of reinforcement, positive and negative reinforcement, have been hypothesized to play a role in this allostatic process. The negative emotional state that drives negati...
